You can clearly see the mark located on your minimap so head to that room and confront the Penanggalan. See here for details. Your reward is a Diamond Bangle which gives a nice boost to your HP.
Other than the usual enemies, there is nothing left to do here so return to the fifth floor. Just note that two Varcolaci will replace the Penanggalan at the same location after it’s dead.
